About the project

This systematic literature review investigates whether sleep deprivation during adolescence increases vulnerability to addiction, and whether dysregulation of the dopamine reward system explains this relationship. Using a structured PubMed search strategy combining terms for adolescence, sleep deprivation, dopamine/reward circuitry, and addiction, restricted to peer-reviewed human studies published between 2000 and May 2026 with participants aged 9-32, the review identifies three studies meeting inclusion criteria, each examining the question through a different lens: experimental designs, longitudinal cohort tracking, and studies of behavioral or chemical stimulus responses. Before synthesizing these studies, the paper builds a detailed neurobiological foundation, explaining the circadian and homeostatic drivers of sleep, the stages of NREM and REM sleep, and the mesocorticolimbic dopamine system linking the ventral tegmental area, nucleus accumbens, amygdala, hippocampus, and prefrontal cortex in reward processing. Synthesizing across the three included studies, the review finds consistent evidence that poorer or reduced adolescent sleep, particularly greater eveningness and shorter weekend sleep duration, is associated with increased substance use and heightened reward sensitivity in subsequent periods, suggesting disrupted communication between dopaminergic reward circuitry and the prefrontal cortex's regulatory control. The review acknowledges significant limitations, including the small number of qualifying studies, heterogeneity in study design and sample size, and unaddressed confounds such as school start times, race, and caffeine use in the underlying studies. The paper concludes that sleep health should be treated as a meaningful, currently underused target for addiction prevention and early intervention in adolescents, given that roughly 90% of Americans with substance use disorders began using substances before age 18, and calls for school-level policy changes and educational programs addressing adolescent sleep.
